OVERALL:
(1) Draw a Direct Plot and a Lineweaver Burke plot from class data. Each graph will have three datasets (Fructose, Mannose and Glucose).
(2) Present a table giving the values obtained from your graphs for Km and Vmax for each substrate. You will need to convert the Vmax values to μmoles/min/1mL assay.
(3) Calculate the relative specificity constants using the Km and Vmax values.
(4) Draw a Lineweaver Burke plot for the xylose inhibition data compared with the uninhibited enzyme for the substrate you used (MANNOSE). Two lines as a minimum on this graph depending on your experimental design.
ANSWER THE FOLLOWING QUESTIONS:
1. Which is the best substrate? Why?
2. Which substrate has the lowest Km?
3. Is xylose an inhibitor? If so what is the type of inhibition?
